Evaluation of Disinfectant Efficacy against Biofilm-Residing Wild-Type Salmonella from the Porcine Industry

چکیده
Salmonella enterica is a causative pathogen of Salmonellosis, zoonosis causing global disease and financial losses every year. Pigs may be carriers contribute to the spread humans food products. persist as biofilms. Biofilms are bacterial aggregates embedded in self-produced matrix known withstand disinfectants. We studied effect glutaraldehyde peracetic acid, two active substances frequently used disinfectant formulations pig industry, on representative biofilm-residing wild-type collected from housings United Kingdom (UK). screened biofilm production strains using microtiter plate (MTP) assay Congo Red Coomassie Blue (CRCB) agar method. Previously published stainless-steel coupon (SSCA), polyvinylchloride (PCA), glass bead (GBA) assays were for disinfection studies. The mean reduction tested met criterion ≥4 log10 CFU at concentration 0.05% with SSCA GBA, 0.005% PCA along 0.5% all three mean. At these concentrations, both disinfectants suitable against Salmonella. When evaluating efficacy disinfectants, biofilms should included, higher concentrations necessary compared planktonic bacteria.

Ruggedness and reproducibility of the MBEC biofilm disinfectant efficacy test.
The MBEC™ Physiology & Genetics Assay recently became the first approved ASTM standardized biofilm disinfectant efficacy test method. This report summarizes the results of the standardization process using Pseudomonas aeruginosa biofilms. Initial ruggedness testing of the MBEC method suggests that the assay is rugged (i.e., insensitive) to small changes to the protocol with respect to 4 factors...

Two-Center Evaluation of Disinfectant Efficacy against Ebola Virus in Clinical and Laboratory Matrices
Ebola virus (EBOV) in body fluids poses risk for virus transmission. However, there are limited experimental data for such matrices on the disinfectant efficacy against EBOV. We evaluated the effectiveness of disinfectants against EBOV in blood on surfaces. Only 5% peracetic acid consistently reduced EBOV titers in dried blood to the assay limit of quantification.
متن کاملImmunization with recombinant Salmonella expressing SspH2-EscI protects mice against wild type Salmonella infection
BACKGROUND Enhancing caspase-1 activation in macrophages is helpful for the clearance of intracellular bacteria in mice. Our previous studies have shown that EscI, an inner rod protein of type III system in E. coli can enhance caspase-1 activation. The purpose of this study was to further analyze the prospect of EscI in the vaccine design. RESULTS A recombinant Salmonella expressing SspH2-Esc...

Methods: The assessments were performed on biofilm attached to dermal tissue using an ex vivo porcine skin explant biofilm model. Its use as a dermal substrate for attachment and the primary source of nutrition provides an alternative cost-effective method that more closely mimics in vivo conditions, with the advantages of an in vitro system, to assess the antimicrobial efficacy of treatments a...
